<b>Biological Strategies Validation. </b>Lysates prepared from LLnV treated U2OS human osteosarcoma cells were incubated in wells coated with Human Total MDM2 Capture Antibody. Unbound material was removed by washing and bound material was solubilized in SDS gel sample buffer. The same lysate and captured proteins were electrophoresed, transferred to a PVDF membrane, and immunoblotted with Human Total MDM2 Detection Antibody. The captured protein corresponds to MDM2 detected in the total cell lysates.

Lysates prepared from U2OS human osteosarcoma +/- LLnV cells were quantified with this DuoSet® IC. The same lysates were also immunoblotted (inset) with a monoclonal detection antibody. The DuoSet® IC results correlate well with the relative amounts of human MDM2 detected by Western Blot.

Western blot analysis on different lysates with NBP3-32584 at 1/1,000 dilution. Lane 1: U-2 OS cell lysate Lane 2: U-2 OS treated with 10μM NUTLIN 3A for 24 hours cell lysate Lysates/proteins at 15 µg/Lane. Predicted band size: 55 kDa Observed band size: 90 kDa Exposure time: 3 minutes; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. NBP3-32584 at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat anti-Mouse IgG [HRP] at 1/50,000 dilution was used for 1 hour at room temperature.

Anti-Mouse IgG Secondary Antibody at 1/5,000 dilution was used for 1 hour at room temperature.

<b>Biological Strategies Validation. </b>Affinity Purified Anti-MDM2 pS185 (Rabbit) is shown to detect a 102 kDa band (arrow) corresponding to phosphorylated mouse MDM2 present in a 293T whole cell lysate. Cells were serum-starved for 24 hours prior to harvest. Approximately 20 ug of lysate was loaded per lane for SDS-PAGE. Untreated cells are shown in lane 1, whereas cells in lane 2 were treated with IGF-1 (100 ng/ml) for 20 min prior to harvest. Follow reaction of antibody with a 1:2000 dilution of HRP Goat-a-Rabbit IgG for visualization.
