TRF-2 (Telomeric repeating-binding factor 2), aka TERF2 and TRBF2, is a ubiquitously expressed protein that is a component of nucleoprotein complex containing duplex 5'-TTAGGG-3' repeats. TRF-2 functions in the regulation of telomere stability. TERF2, like TERF1, contains a Myb-realted DNA binding motif and plays a key role in shield chromosome ends from degradation and end-to-end fusion, as well as preventing activation of DNA damage checkpoints. Mutant TRF-2 studies have shown that the protein is required for the prevention of senescence in primary human cells. Recently, it has been shown that inhibition of TRF2 resulted in apoptosis in a subset of mammalian cell types. TRF-2 is overexpressed in a number of human tumors, so overexpression of TERF2 is thought to be oncogenic.
