Description

IRE-1 alpha is a transmembrane protein that has both intrinsic serine-threonine kinase and endoribonuclease activities. IRE1 alpha is critical for altering gene expression in response to endoplasmic reticulum-based stress signals.

Specifically, IRE1 alpha senses unfolded proteins in the lumen of the endoplasmic reticulum via its N-terminal domain, which leads to enzyme auto-activation. The active endoribonuclease domain splices XBP1 mRNA to generate a new C-terminus, converting it into a potent unfolded-protein response transcriptional activator and triggering growth arrest and apoptosis. Null mutants are viable, but IRE1 is essential for viability under stress conditions that cause unfolded proteins to accumulate in the ER.
