The notch proteins are single-pass membrane receptors controlling a highly conserved signalling pathway. In vertebrates, the notch family comprises four transmembrane receptors encoded by four individual genes (NOTCH1 - 4). In 1999, the notch signalling cascade was shown to play multiple roles in cell fate decisions, controlling the cellular response to intrinsic or extrinsic signals associated with developmental programming. Notch signalling is an important initiator of cellular differentiation and proliferation, as well as apoptosis, and a basic developmental tool for morphogenesis and organ formation.

Notch proteins are synthesised in inactive form in the endoplasmic reticulum, being activated by S1 cleavage in the Golgi apparatus. The active receptor is a hetero-oligomer spanning the plasma membrane. It has a large extracellular portion which associates with a smaller structure composed of an intra/extracellular region, forming a single transmembrane-pass.

Various NOTCH antibody studies have shown that the receptor protein is normally only triggered by direct cellular contact, with activation achieved by extracellular binding of transmembrane ligands. This induces S2 cleavage of the intracellular domain, which then translocates to the nucleus, targeting the DNA binding protein RBP-Jκ/CBF1 and activating transcription of notch target genes. In 2001, a study by Oswald Tauber, et al. showed evidence that p300 acted as a transcriptional co-activator for the notch-1 pathway.
