A versatile member from the WNT protein family

The Wnt gene family is made up of structurally related genes encoding secreted signaling proteins. These proteins have been implicated in oncogenesis and developmental processes, such as cell fate regulation and embryogenesis patterning. There is high evolutionary conservation among species for the Wnt5A protein. Rao’s group from China used the Wnt5A antibody in their immunohistochemical expression survey on a wide variety of renal epithelial neoplasms in hopes of determining if Wnt5A is a useful immunohistochemical marker that allows a definitive differential diagnosis between renal oncocytomas and other subtypes within the spectrum of renal cell carcinomas (RCCs)1. Some interesting neuroscience studies detailing synaptic plasticity and chronic pain were performed with the Wnt5A antibody in HIV patients2.

These researchers compared levels of ligands, receptors, and downstream effector molecules of the Wnt signaling pathway to create a signaling model for the spinal cord dorsal horn (SDH) in both pain-positive and pain-negative HIV patients. Siar’s group used the Wnt5A antibody to examine the role of Wnt signaling in odontogenesis and progression of ameloblastoma, an epithelial neoplasm with odontogenic origin3.  With the Wnt5A antibody, they were able to carefully profile the differential expression of Wnt ligands in different ameloblastoma subtypes – they found that while the canonical Wnt pathway drives general odontogenesic signaling, the non-canonical Wnt pathway is differentially involved in tumorigenesis. The Wnt5A antibody also enabled Olivares-Navarrete et al to examine non-canonical, calcium-dependent Wnt signaling in osteoblasts allowed to mature on microstructure titanium surfaces during implant osseointegration4.
