![Immunoprecipitation: beta-III Tubulin Antibody (2G10) [NBP2-37816] - Analysis of beta-3 Tubulin was performed using SHSY5Y whole cell lysate. Antigen-antibody complexes were formed by incubating 300ug of lysate with 5ug of a beta-3 Tubulin monoclonal antibody overnight on a rocking platform at 4C. The immune complexes were captured on 50ul Protein A/G Agarose, washed extensively, and eluted with 5X Lane Marker Reducing Sample Buffer. SHSY5Y cell lysate (30ug) was loaded as a positive control (left lane). The sample was resolved on a 4-20% Tris-HCl polyacrylamide gel, transferred to a PVDF membrane, and blocked with 5% BSA/TBS-0.1%Tween for at least 1 hour. The membrane was probed with a beta-3 Tubulin monoclonal antibody at a dilution of 1:1000 overnight rotating at 4C, washed in TBST, and probed with Clean-blot IP Detection Reagent at a dilution of 1:1000 for at least 1 hour. ![Immunohistochemistry: alpha Tubulin Antibody (PSH02-95) [NBP3-33140] - Immunohistochemical analysis of paraffin-embedded rat brain tissue with Rabbit anti-alpha Tubulin antibody (NBP3-33140) at 1/1,000 dilution. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-33140) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Cells were then incubated with Rabbit anti-alpha Tubulin antibody (NBP3-33140) at 1/100 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (iFluor™ 488) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI. Beta tubulin (red) was stained at 1/100 dilution overnight at +4℃.
