Green Fluorescent Protein
GFP (green fluorescent protein) is a fluorescent protein produced by the crystal jellyfish, Aequorea victoria; which produces glowing points of light around its umbrella's margins through bioluminescence reactions in photogenic cells. GFP gene has been isolated and has emerged as a useful tool for making expressed proteins fluorescent by generating chimeric genes composed of those of GFP/its different colour variants linked to genes of target proteins. GFP is now a widely used reporter protein in eukaryotic/prokaryotic gene expression studies, and as a protein tag in tissue culture as well as multicellular organisms for localizing and studying protein movement/dynamics in cells or tissues. GFP epitope tags are useful for labeling/detection of proteins using various techniques including immunodetection methods and GFP does not affect the tagged protein's biochemical properties. GFP absorbs light maximally at 395 nm with an emission spectrum that peaks at 509 nm and has become a very useful tool as a fusion protein that reports gene expression, traces cell lineages and defines subcellular protein localizations.
