Background

Dendritic cells are potent antigen presenting cells that possess the ability to stimulate naive T cells. They are critical for the induction of T cell responses resulting in cell mediated immunity. Within different tissues, dendritic cells differentiate and become active in the taking up and processing of antigens, and their subsequent presentation on the cell surface linked to major histocompatibility (MHC) molecules. Upon appropriate stimulation, dendritic cells undergo further maturation and migrate to secondary lymphoid tissues where they present Ag to T cells and induce an immune response.

  1. I have a question regarding the antibody AP-MAB0801: The product description is very unspecific, saying it recognizes a surface marker of mouse DCs. We would like to know which surface marker this antibody recognizes, so we know which subsets of DCs will be stained.
    • This clone detects the Dendritic Cell Inhibitory Receptor 2 on mouse DC cells.
  2. Does the antibody stain DCs in mouse spleen sections?
    • Yes
  3. Which DCs does it stain?
    • NB110-85474 will stain all mouse dendridic cells.
  4. Could you let us know which kind of dendritic cells from mouse have been used to generate the antibody? We want to use the antibody for staining of intravascular dendritische cells of the central nervous system.
    • The lab has informed me that purified Spleen dendritic cells were used as immunogen for this clone. The lab also informed me that the immunogen cells were DC enriched cells from spleen cell suspension and for antibody binding 100% pure cells are not required.
