Member of 1-acylglycerol-3-phosphate-O-acyltransferase (AGPATs) family, AGPAT6 (glycerol-3-phosphate acyltransferase 4) is an enzyme essential for biosynthesis of glycerolipids and triacylglycerol in eukaryotes, as well as for catalyzing the conversion of lysophosphatidic acid to phosphatidic acid. It esterifies acyl-group from acyl-ACP to the sn-1 position of glycerol-3-phosphate, which is a required step in glycerolipid biosynthesis. AGPAT6 is active against both saturated as well as unsaturated long-chain fatty acyl-CoAs and is inhibited by N-ethylmaleimide (NEM). Its HXXXXD motif is critical for its acyltransferase activity and constitutes the site for binding of glycerol-3-phosphate's phosphate moiety. Localized in ER membrane as multi-pass membrane protein, AGPAT6 is ubiquitously expressed with relatively high expression levels in skeletal muscle, heart, mammary epithelium and testis. AGPAT6 contributes to the production of milk fat and has been considered a candidate gene for regulating milk composition. Mouse model deficient in AGPAT6 exhibit weight loss and resistance to both diet-induced as well as genetically induced obesity.
