Background

Toll-like receptors (TLRs) are signaling molecules that recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. These proteins act through adaptor molecules such as MyD88 and TIRAP to activate various kinases and transcription factors. TLR11 is one of three mouse TLRs that lack a human ortholog. It is activated specifically by uropathogenic bacteria, and mice lacking TLR11 showed a much greater susceptability to uropathogenic infections, indicating a potentially important role for TLR11 in preventing infections in the urogenital system.

  1. I have purchased TLR11 stable cell line from your company and cells are now in culture. In case of other cell lines, transfected with TLRs, a special medium had to be prepared for stimulation of these cells. In the available datasheet, the listed publications and also in other literature, I can't find any information if the stimulation has to be performed in normal growth medium or in a special one. Can you give any further information regarding this problem?
    • We generally use normal culture media without selectable antibiotics when we analyze our TLR-transfected stable cell lines (such as ligand stimulation, signaling inhibitor test, agonist/antagonist test, etc). However, when you maintain the cell lines, you should not omit the selectable antibiotics in the culture media.
