mCherry represents an engineered derivative of a family of proteins initially isolated from a group of sea organisms called Cnidarians (jelly fish, sea anemones and corals). The prototype for these fluorescent proteins is GFP which was originally isolated from jellyfish Aequoria Victoria and when expressed in-vitro or in-vivo using its gene, GFP protein fold correctly with retention of its capability to deliver strong fluorescence. Engineered forms of GFP and its relatives have been developed to monitor several biological proresses such as Ca2+ levels, protease activation and a variety of other processes in real time. mCherry protein was derived from DsRed, a red fluorescent protein from so-called disc corals of Discosoma species. DsRed is similar in size and other properties to GFP, but, produces a red rather than a green fluoresnecce and its engineered monomeric form has several applications including FRET (Forster Resonance Energy Transfer). mCherry was engineered with further multiple cycles of mutation, directed modification and evolutionary selection, and it is also a monomeric form with an excitation maximum at 587 nm and an emission maximum at 610 nm.
