Application Notes
This antibody has been assayed against 1.0 ug of Asparaginase in a standard capture ELISA using Peroxidase Conjugated Streptavidin and ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:3,200 to 1:12,800 of the reconstitution concentration is suggested for this product.

Notes

This antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Biotin, anti-Rabbit Serum as well as purified and partially purified Asparaginase [E.coli]

Store vial at -20C or below prior to opening. This vial contains a relatively low volume of reagent (25 ul). To minimize loss of volume dilute 1:10 by adding 225 ul of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20C or below after dilution. Avoid cycles of freezing and thawing.

Background

Asparaginase is an enzyme purified from E. coli and Erwinia carotovora. It acts by deaminating extracellular L asparagine, an amino acid that appears to be essential for protein synthesis by some tumour cells which are unable to synthesise asparagine. Asparaginase from Erwinia carotovora is serologically and biochemically distinct from asparaginase from E. coli, although its antineoplastic activity and toxicity is similar. Asparaginase is usually considered to be cell cycle phase nonspecific, but it may block some cells in G1 or S phase. Asparaginase reduces cellular and humoral immunity. E.coli contains two L asparaginase isoenzymes: L asparaginase I, a low affinity enzyme located in the cytoplasm, and L asparaginase II, a high affinity secreted enzyme.
