Western blot shows lysates of human brain tissue, Daudi human Burkitt lymphoma, and A549 human lung carcinoma cells. PVDF membrane was probed with 1 µg/mL of Human PYK2/FAK2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4589) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for PYK2/FAK2 at approximately 116 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

PVDF membrane was probed with 0.5 µg/mL of Rat Anti-Human Phospho-FAK (Y397) Monoclonal Antibody (Catalog # MAB4528) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Phospho-FAK (Y397) at approximately 150 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Western blot shows lysates of HEK293T human embryonic kidney parental cell line and FAK knockout HEK293T cell line (KO). PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human/Mouse/Rat FAK Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4467) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for FAK at approximately 135 kDa (as indicated) in the parental HEK293T cell line, but is not detectable in knockout HEK293T cell line. GAPDH (Catalog # AF5718) is shown as a loading control. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Western blot shows lysates of Raji human Burkitt's lymphoma cell line and Jurkat human acute T cell leukemia cell line untreated (-) or treated (+) with 1 mM Pervanadate (PV) for 30 minutes and 10 µg/mL Mouse Anti-Human CD3e Monoclonal Antibody (Catalog # MAB100) for 15 minutes. PVDF membrane was probed with 0.5 µg/mL of Mouse Anti-Human Phospho-PYK2/FAK2 (Y402) Monoclonal Antibody (Catalog # MAB6210) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). Specific bands were detected for Phospho-PYK2/FAK2 (Y402) at approximately 105-115 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
