Brentuximab Antibody (2262C) [Unconjugated]

Western blot shows Centuximab (negative control), Rituximab (negative control), Brentuximab, and Gemtuzumab (negative control) biosimilars. PVDF membrane was probed with 0.5 µg/mL of Anti-Brentuximab (Idiotype) Monoclonal Antibody (Catalog # MAB9584) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (HAF008). A specific band was detected for Brentuximab at approximately 240 kDa (as indicated). This experiment was conducted under non-reducing conditions and using Immunoblot Buffer Group 1.

Jurkat human cell line was stained with anti-CD30/TNFRSF8 (Brentuximab, Catalog # MAB9576) then Anti-Brentuximab (Anti-Idiotype) Monoclonal Antibody (Catalog # MAB9584, filled histogram) or normal rabbit IgG isotype control antibody (MAB1050, open histogram), followed by PE-conjugated anti-rabbit IgG secondary antibody (F0110).

Biotinylated biosimilars were serially diluted and incubated with Anti-Brentuximab (Anti-Idiotype) Antibody (Catalog # MAB9584, green line) coated at 1 μg/mL on a 96-well polystyrene plate. No cross-reactivity with Rituximab (black line) or biotinylated IgG control (orange line) was observed.

A panel of biosimilars was coated at 1 μg/mL in 96-well polystyrene plates. Anti-Brentuximab (Anti-Idiotype) Antibody (Catalog # MAB9584, 333 ng/mL) detected Brentuximab. No cross-reactivity with Cetuximab, Rituximab, Trastuzumab, Adalimumab, Gemtuzumab, Ibritumomab, Tositumomab, Solanezumab, Teprotumumab, and Aducanumab was observed.
