The relative AGO protein levels were detected by western blot 56 hours post siAgo transfection, at the time point when Ago2/eIF2C2 ablated cells were harvested to evaluate luciferase activities. GAPDH was included as loading control. The intensity of protein bands was quantified by ImageJ software (NIH, USA). All experiments were performed at least twice.
