Description

SIT is a nbsp40 kD disulfide-linked protein also known as SHP2-interacting transmembrane adaptor protein. SIT is a membrane protein containing a very short extracellular domain (18 amino acids). SIT is expressed in T cells, but is not expressed in monocytes. SIT is a signaling adaptor molecule that negatively regulates T cell activation through SHP-2 interactions. The SIT protein is phosphorylated by Src kinases and has been shown to associate with SHP2, Csk, Zap70, Syk, Fyn, Lck, and Grb2. SIT is also heavily glycosylated and treatment with endoglycosidase has been shown to reduce the apprarent molecular weight to approximately 20 kD. The SIT-40 antibody recognizes the intracellular region of human SIT and has been shown to be useful for Western blotting.
