TUNEL Apoptosis Detection Kit provides all the reagents for
the detection of DNA fragmentation among vascular cells and tissue sections. It
generates an intense blue staining in cells with DNA fragmentation, easily visualized
against a pale red counterstain. In addition, Cytonin, a non-lipophilic detergent based
buffer optimized for the permeabilization of cells prior to labeling, and Blue Streptavidin Diluent,
a proprietary blocking reagent that contributes to the low background obtained with
Tunel Apoptosis Detection kit are provided.

Background
Apoptosis is characterized by a number of intracellular phenomena such as membrane blebbing, chromatin condensation and nuclear DNA fragmentation. Detection of nuclear DNA fragmentation is a widely accepted method to assay for apoptosis and can be performed in situ by incorporating labeled nucleotides onto the free 3' OH ends of DNA fragments using a terminal deoxynucleotidyl transferase enzyme (TdT). Terminal dUTP nick-end labeling (TUNEL) allows monitoring of apoptotic events among cells and within tissue sections. The incorporation of biotinylated nucleotides allows chromosomal DNA fragmentation to be visualized by binding streptavidin-horseradish peroxidase followed by reaction with TACS Blue Label(TM) to generate a dark blue precipitate. When viewed under a standard light microscope apoptotic cells are clearly distinguished by dark blue staining.
