Description | Tissue Clearing Pro-Organoid is a rapid, easy-to-use, reversible and non-destructive kit, which allows effective tissue clearing and staining of organoids, 3D cell cultures and microtissues. This organoid tissue clearing kit allows 3D cell cultures up to 500 µm thick to be cleared in minutes, and is compatible with fluorescent protein, immunolabeling and small molecule staining techniques, as well as being suitable for use with all standard dyes and buffers. Following 3D imaging, Tissue Clearing Pro-Organoid can be reversed for follow-up 2D histology staining. The kit comes with a getting started guide and a more comprehensive tissue clearing guidebook, providing detailed protocols. Tissue Clearing Pro-Organoid kit contains 30 mL of Tissue Clearing Pro-Organoid clearing reagent plus the 4 buffer solutions required to process 3D cell cultures for immunolabeling and is enough to process three 96-well plates of material. A wide range of IHC validated antibodies are available from R&D Systems and Novus Biologicals; optimization of antibody incubation conditions for use with the Tissue Clearing Pro-Organoid technique is recommended. Application Notes | The Tissue Clearing Pro-Organoid kit is designed specifically for use with 3D cell cultures, organoids and microtissues. It is able to clear samples with minimum changes to morphology and without compromising the sensitivity of detection with almost any fluorophore. With this easy-to-use protocol, 3D cell cultures up to 500 um in thickness can be cleared within minutes, without using any special instrument or equipment. The clearing workflow is compatible with most fluorophores including fluorescent proteins, which can be detected with typical fluorescent imaging instruments such as wide-field, confocal, and light sheet microscopes, and high-content analyzers. The Tissue Clearing Pro-Organoid technique is strong enough to adequately clear tissue for 3D fluorescent imaging, but not so harsh as to change the overall tissue morphology. Minimum morphological changes such as shrinkage or contraction have been observed. For precious samples, the clearing can be reversed, and tissue can be processed for histology studies such as H&E staining. |
