Description
The performance of HRP conjugates diminishes over time. There are a number of factors that contribute to loss of performance including:
  • Inactivation of HRP.
  • Instability of the bond which links HRP to the antibody.
  • Microbial attack.
  • Denaturation of the antibody.
  • Loss of performance accelerates with increasing temperature and with increasing dilution of the conjugates.
For these reasons HRP conjugates are often kept in a concentrated form at 4oC.

The conjugate stabilizer/diluent is a proprietary multi-component reagent system that protects antibody-HRP conjugates from all of the negative factors noted above, thus ensuring the best possible performance in experiments performed at room temperature. Moreover, the ability to store HRP conjugates at working dilutions eliminates waste and improves consistency from experiment to experiment.

In addition to its proprietary enzyme stabilizers, it contains antimicrobial agents as well as blockers to enhance signal to background in immunoassay applications.
Specificity
Even some of the best known stabilizers perform either poorly or only modestly in standard accelerated stability trials at 37C. LifeXtend HRP stabilizer/diluent affords the greatest protection and can substantially extend the shelf life of HRP conjugates stored at 4C or at room temperature.

CommentsWe use an immobilized antibody competitive EIA for running hormone assays. We coat the plate with our antibody & load the next day - we add our HRP:hormone conjugate at that time. We make up working stock of HRP conjugates (~1:50 or 1:100 refrigerated) in small batches because most of them get weaker over a period of time (1-2 months). We make our loading dilutions at time of plate loading. We are trying to stabilize the refrigerated working stock for a longer period of time. So far, I have used the LifeExtend to make up our progesterone HRP conjugate. It has only been 2-3 weeks so we will be monitoring it over a longer period of time to determine the shelf life of HRP conjugate. At this time we can say that it has no detrimental effects at all on the assays (this was unclear when I asked about antigen conjugates versus antibody conjugates). As I make up more HRP stocks (PdG, estrone glucuronide, testosterone, cortisol, corticosterone, etc), I will be using the LifeExtend. Hopefully, this will extend the shelf life of our working stocks so that we can make larger batches at a time & not compromise the integrity of our original frozen stock through repeated freeze-thaws to make small batches.
