Specificity
It reacts with Bromodeoxyuridine (BrdU) in single stranded DNA (produced by partial denaturation of double stranded DNA), BrdU coupled to a protein carrier, as well as free BrdU. BrdU is a thymidine analog, incorporated into cell nuclei during DNA synthesis prior to mitosis. Antibody to BrdU is helpful in detecting S-phase cells, providing useful information on the aggressiveness of tumors.

Background

BrdU (Bromodeoxyuridine, BUdR, BrdUrd) is a synthetic nucleoside that is an analogue of thymidine. It is commonly used in the detection of proliferating cells in living tissues (1). BrdU can be incorporated into the newly synthesized DNA of replicating cells (during the S phase of the cell cycle), substituting for thymidine during DNA replication. An antibody specific for BrdU can then be used to detect the incorporated chemical, thus indicating cells that were actively replicating their DNA. Binding of the Brdu antibody requires denaturation of the DNA.

  1. I have been using your BrdU antibody for some time now and saw that you have discontinued it. I was wondering if you had any alternative antibodies that would also work? it is antibody NB500-169 lot# 1706-5
    • We recommend this product: NB500-235​
  2. How can I use this antibody on mouse tissue and be confident there is no cross-reactivity?
    • Our BrdU antibody with catalogue number NBP2-29414 has been validated for IHC with paraffin-embedded samples from human and mouse, however its reactivity with other species and in other applications has not yet been tested. When performing IHC with paraffin-embedded samples we routinely include an isotype control as well as a no primary antibody control. The isotype control is used to differentiate non-specific background signal from the specific antibody signal, whilst the no primary antibody control is used to check that the secondary antibody is not binding to the sample. You can see our mouse IgG1 kappa isotype controls. Additionally, I am sure that you are aware of the need for a mouse-on-mouse block when using mouse antibodies with mouse tissue, however please find here a link to a good source of mouse-on-mouse regents, which you may find helpful
