Recognizes conjugated Acrolein. Using a conjugate Acrolein Protein, specificity was performed with an ELISA test by competition experiments with the following compounds. Acrolein-BSA (ratio 1:1), Malondialdehyde-BSA (1:55), Glyoxal-BSA (1:>50,000), Formaldehyde-BSA (1:>50,000), Glutaraldehyde-BSA (1:> 50,000)

Details:
Acrolein antibody used in Dot Blot application for detecting the accumulation of acrolein-lysine adducts in the spinal cords of mice following nasal exposure to acrolein. Fig 3 shows the representative dot blots for control, acrolein, and sham groups.

  1. Could you please let us know the Molecular Weight of NB200-556 for WB?
    • Acrolein is a small organic compound (it is an unsaturated aldehyde; not a protein). NB200-556 recognizes acrolein-modifed proteins; the molecular weight will depend on the MW of the protein to which the acrolein is conjugated to (protein modification by acrolein can be ascribed to the formation of acrolein-lysine and/or acrolein-histidine adducts that possess a carbonyl function). This also applies to the size of the protein.
  2. I have recently purchased Acrolein antibody (NB200-556) and used for western blotting. I have got two bands one is very faint (50Kda) and second nice band at 5-15 Kda. I was wondering at what location band for Acrolein (mol wt.) should show up in western blotting. Should I consider faint band (50KDa) also ??
    • Acrolein is a simple aldehyde. Detecting this molecule alone is impossible on Western blot as it is far too small. If it is contained in your protein or molecule of interest, it will be detected at the molecular weight of the protein or molecule. Because of this, it is impossible to give you a size to look for as everyone's samples will be different. Since this is a very unique antibody, I would treat any band that shows up when detecting with NB200-556 as being Acrolein positive.
