Adeno-associated virus (AAV) is a small, non-enveloped single-stranded DNA virus that requires a helper virus to replicate. The AAV genome consists of a rep and cap gene in which the rep gene encodes for AAV’s non-structural proteins, and the cap gene encodes the structural proteins (VP1, VP2, VP3) that form the viral capsid.
