Presenilin (PS1/PS2) complexes are responsible for the intramembrane gamma-secretase cleavage of the beta-amyloid precursor protein. Presenilin enhancer 2 (PEN2) is an essential cofactor involved in physically interacting with PS1 and is necessary for gamma-secretase activity. Endogenous PEN2 preferentially interacts with PS1 holoprotein. Down-regulation of PEN2 expression abolishes the endoproteolysis of PS1 and overexpression promotes the production of PS1 fragments.
