Description
Phosphatidylinositol 3 Kinase (PI3 Kinase) is a heterodimeric enzyme that phosphorylates the D3 position of the inositol ring of PtdIns, PtdIns-4-P and PtdIns-4,5-P2. The production of D3 phosphorylated polyphosphoinositides defines a novel phosphatidylinositol signaling pathway. The regulatory 85kD subunit of PI3 Kinase consists of two SH2 domains, two proline rich sequences and an SH3 domain. The amino terminus of the p110 catalytic subunit associates with the inter SH2 domain of the 85kD subunit. PI3 Kinase has been shown to be essential for PDGF induced mitogenesis and transformation by polyomavirus middle T antigen. In addition, PI3 Kinase is known to be activated by a large number of mitogens and oncogenes and the enzymatic activity is conserved across species and down the phylogenic tree to yeast. Three isoforms have been cloned to date, p110a, p110b and p110g. The significance of the different isoforms is not yet understood.
Phosphorylates PtdIns. PtdIns4P and PtdIns(4.5)P2 with a preference for PtdIns(4.5)P2.
