1. Plate cells at 2 x 10^6/ml in regular cell culture medium.
2. Add inhibitor peptide to a final concentration of 100 uM for 12 hrs.
Example: 40 ul of inhibitory peptide or control peptide stock solution, added to a final volume of 2 ml cell culture media, will give a final concentration of 100 uM.
3. Prepare nuclear extracts and check for the presence of NF-kB DNA-binding activity by EMSA (Figure 1). Nuclear extracts can be prepared as described by Bharati A, et al., or using nuclear extraction kit
