Do you know the size of the inhibitors in this cocktail? I am going to denature the proteins, so if they are susceptible to denaturation I can forget about them? Or should I use a spin column to remove low molecular weight compounds?
- These inhibitors are all < 10 kDa in size, so if you don't want to buy a large quantity, you can definitely use a spin column with a 10 kDa molecular weight cutoff to remove them. I feel that this is the better strategy. This is because, without protease inhibitors, there will be both denaturation and degradation of the lysate protein. If you are performing protease studies, it is better to start with an intact full-length protein.

I want to test different proteases on a variety of substrates, and thought universal whole tissue lysate as a good choice. Unfortunately it contains a cocktail of protease inhibitors. Is it possible to get some without these inhibitors?
