Description
The TLR3 / ISRE / LUCPorter™ Reporter Cell Line can be used for studying the IRF3-dependent induction of type I interferons as well as screening of agonists/antagonists/signaling inhibitors related with IRF3-dependent TLR3 activation. Functional activity of the cell line has been validated by TLR3 ligand dose assay (Figure 1), in which upon activation by poly (I:C), TLR3 quickly initiates TRIF-dependent signaling pathway and mediates nuclear translocation of IRF3 that induces ISRE activity.

The TLR3 / ISRE / LUCPorter™ Reporter Cell Line is a stably co-transfected cell line which expresses full-length human Toll-like receptor 3 (TLR3) and an optimized Renilla luciferase reporter gene (RenSP) under the transcriptional control of the interferon stimulated response element (ISRE).

Complete Growth Medium: DMEM with 4.5 g/L glucose + 10% FBS + 4 mM L-glutamine + 1 mM sodium pyruvate + 100 units/ml penicillin + 0.1 mg/ml streptomycin + 10 ug/ml blasticidin + 3 ug/ml puromycin.

Notes

Assume all cultures are hazardous since they may harbor latent viruses or other organisms that are uncharacterized. The following safety precautions should be observed.
- Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
- No eating, drinking or smoking while handling the TLR3 reporter cell line.
- Wash hands after handling the TLR3 reporter cell line and before leaving the lab.
- Decontaminate work surface with disinfectant or 70% ethanol before and after working with the cell line.
- All waste should be considered hazardous.
- Dispose of all liquid waste after each experiment and treat with bleach.

  1. The customer said that HEK293 cells weave what expresses TLR with what does not express TLR. Is the HEK293 of this product, NBP2-26292, derived from the HEK293 cell of which does not express TLR?
    • NBP2-26292 is made with standard HEK293 cells. Hence any endogeneous TLRs would presumably be present; however, we have not done extensive evaluations to determine if the transfection/stable selection itself has caused any alterations in endogeneous TLR expression. Nor have we extensively evaluated endogeneous TLR expression. However, on the TLR stable cell line data sheets there is some data showing TLR expression in transfected versus untransfected cell lines.
