• Description: Inorganic phosphate (Pi) is a product of many enzymatic reactions, for example, those catalysed by phosphatases and ATPases. Measurement of released Pi allows enzyme activity to be determined, but contamination of enzymes or buffers by Pi can lead to unacceptably high assay backgrounds.
  
  PiBind resin provides a quick and easy way to remove contaminating Pi from buffers. The resin works over a broad range of pH values and is unaffected by many commonly used buffer additives. PiBind resin can also be used to remove Pi from protein samples (e.g. tissue extracts). This method is quicker than dialysis and unlike desalting does not necessarily lead to a significant dilution of the sample.

Background

PiBind resin has an extremely high affinity for inorganic phosphate, and is used to eliminate contaminating Pi from buffer solutions to reduce background signals in phosphatase or ATPase assays. PiBind resin may also be used to remove Pi from cell and tissue extracts and is a rapid alternative to desalting or dialysis.

This product is for research use only and is not approved for use in humans or in clinical diagnosis. Support products are guaranteed for 6 months from date of receipt.

1. How do I determine how much PiBind resin to use?
   • Without adding the ATP, carry out the assay as described in the protocol.You should then be able to quantify the amount of phosphate contamination. Once you have done this, you can calculate the amount of PiBind that would be necessary to purify the sample.
