Background

Noradrenaline (norepinephrine) is extracted by using a cis-diol-specific affinity gel, acylated and then converted enzymatically. The competitive ELISA kit uses the microtiter plate format. The antigen is bound to the solid phase of the microtiter plate. The derivatized standards, controls and samples and the solid phase bound analytes compete for a fixed number of antibody binding sites. After the system is in equilibrium, free antigen and free antigen-antibody complexes are removed by washing. The antibody bound to the solid phase is detected by an anti-rabbit IgG-peroxidase conjugate using TMB as a substrate. The reaction is monitored at 450 nm. Quantification of unknown samples is achieved by comparing their absorbance with a reference curve prepared with known standard concentrations.

  1. I would like to measure adrenergic receptors (alpha and beta) in lympocytes and platelets, respectively; can you help? I would like to measure beta and alpha adrenergic receptors in rat lympocytes and platelets. Quotes for ELISA kits to measure insulin, in 250 samples, ELISA kit to measure norephinephrine.
    • We do have an ELISA kit for norepinephrine (catalog # KA1891). Unfortunately, we do not have experience in (or knowledge of) measuring the density of adrenergic receptors; we suggest you consult the literature for suitable methods. If you then decide you would like to order a particular product from us, we would love to hear from you again.
  2. I want to measure noradrenaline level in mouse plasma. please let me know whether the product (KA1891) is reactive to mouse.
    • The norepinephrine ELISA kit KA1891 is a product we distribute for a Taiwanese company called Abnova. This particular kit is an enzyme immunoassay for the quantitative determination of Norepinephrine in plasma and urine. Since norepinephrine is a hormone and not a protein, the molecule is species independent. Therefore, this kit will meet your needs. I will include a link to the protocol included with the kit for your reference
  3. Will this work for bovine norepi?
    • For kit KA1891, since the ELISA target is a neurotransmitter and hormone, it will not be species specific, but specific for the compound itself. Therefore, this kit can be used with a variety of species. The only restriction for this kit will be the sample type. Thus far, it has only been tested with Plasma and Urine.
  4. Can I dilute the sample with NaOH to increase the pH?
    • Yes, pH adjustment by addition of NaOH can be an option in this case. A pH around 3 or 4 would be ideal because that would be normal for urine. The pH of the standards is around 1, so this might also work. But it might denature the proteins of the urine, so a higher pH would be better.
  5. Can this kit be used with rat serum/plasma?
    • Rat serum and plasma samples can be tested with the KA1894, KA1891 and KA1877. There is no species cross-reactivity between rat samples and the antisera of our assays.
  6. What is the difference between the Quantikine and Duoset ELISA kits?
    • Our Quantikine kits are fully optimized and validated for the sample types listed on the product specific webpage and datasheet, as this can vary between kits. Each kit is supplied ready to use with one pre-coated 96-well plate, a detection antibody directly conjugated to HRP, and all other necessary reagents. These kits are ideal for researchers who want the convenience of a ready to use and optimized ELISA product. Some of these kits are also available prepackaged in larger 6 and 50 plate sizes.Our DuoSet Kits, in contrast, are ELISA development kits containing the capture and detection antibody, the mass-value calibrated standard, and streptavidin-HRP to prepare approximately 5 or 15 plates. Ancillary reagents will need to be used/purchased, and for most kits, we will recommend one of our Ancillary Reagent Kits, which contain the reagents we use ourselves in-house. DuoSet kits are validated only for cell culture supernatant samples and therefore require further development and validation for accurate measurement in more complex samples such as serum and plasma.
