Details:
MRG-1 antibody used for IHC-Fr on C. elegans wild-type and rfp-1(ok572) mutant gonads - five to eight minutes/RT fixation in 3% paraformaldehyde , post-fixation in 100% methanol -20C ON , 1 hour/RT blocking in 30% goat serum, primary antibody used at 1:1000 with ON 4C incubation (Fig 7C). IHC-Fr also performed on N2 (wild-type), mrg-1(RNAi) and mrg-1(tm1227) M+Z- animals (Figure S2B). Antibody also used for WB on N2 (wild type) C. elegans as well as the mutants pas-5(oz237) and rfp-1(ok572), and on C. elegans treated with lactacystin, a proteasome inhibitor (Fig 7A, 7B). WB also performed on N2 wild type in comparasion to MRG1-RNAi, MRG1 M+Z- and MRG1 M+Z+ (Figure S2A). Antibody also used for IP application on wild-type animals treated or not with Lactacystin (Figure S4).
