hNPCs are derived from integration-free, induced pluripotent stem (iPS) cells under fully defined neural induction conditions. The hNPCs express typical markers of cerebral cortical neural stem and progenitor cells e....g. Pax6 and Foxg1, and spontaneously form polarized neural tube-like rosette structures when plated as a monolayer in culture. Additionally, hNPCs are capable of generating a spectrum of cortical neurons that are electrically active and have the ability to form functional synapses and circuits in vitro. Cerebral cortical neurons are implicated in numerous neurological diseases including Alzheimer's disease, autism, epilepsy and stroke. Despite strong interest in these diseases, high quality human in vitro cell models for cortical neurophysiology were unavailable until now.
