Western Blot: MyoD Antibody (5.8A) [NB100-56511] -  Analysis for MyoD expression in various small round cell tumor lines using 1 ug/ml anti-MyoD mAb. The antibody only reacts with a band of approx. 45 kD in the rhabdomyosarcoma cell line (Rh30, lane 1) but was negative against the primitive neuroectodermal (PFSK-1A, lane 2), lymphoma (EB2, lane 3), neuroblastoma (SK-N-SH, lane 4), and Ewing's sarcoma (SJSA-1, lane 5) cell lines.Immunocytochemistry/Immunofluorescence: MyoD Antibody (5.8A) [NB100-56511] - RD cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-MyoD1 (5.8A) NB100-56511 at a 1:200 dilution overnight at 4C and detected with and anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was counterstained with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: MHC Class I Antibody (OX18) [NB120-6405] - PC-12 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-MHC Class I (OX18) NB120-6405 at a 1:100 dilution overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.Immunohistochemistry-Paraffin: MHC Class I Antibody (OX18) [NB120-6405] -  IHC analysis of formalin fixed paraffin-embedded (FFPE) rat brain cerebellum using MHC Class I (OK18) antibody at 1:200 on a Bond Rx autostainer (Leica Biosystems). Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Endothelial staining was observed. Staining was performed by Histowiz.

The membrane was probed with 2.0 ug/ml anti-AKT1 (NBP2-01725) and 2 ug/ml pS473 AKT1 in 1% BSA in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence. This antibody generated a diffused cytoplasmic staining of phosphor-AKT (Ser-473) in the cancer cells as well as the stromal cells. Some cancer cells depicted nuclear stianing also. Staining was performed by Histowiz.
