![Western Blot: IL-6 Antibody [NBP1-77894] - Shows detection of recombinant mouse IL-6 raised in E.coli. ![Immunohistochemistry: IL-6 Antibody [NBP1-77894] - Shows nuclear positivity of islets of Langerhans (brown staining) and cytoplasmic staining in mouse pancreas at 10x and 20x (B & C). Staining was performed on Leica Bond system using the standard protocol. Formalin fixed/paraffin embedded tissue sections were subjected to antigen retrieval with E1 (Leica Microsystems) retrieval solution for 20 min and then incubated with rabbit anti-mouse IL-6 antibody at 1:50 dilution for 60 minutes. Biotinylated Anti-rabbit secondary antibody was used at 1:200 dilution to detect primary antibody. The reaction was developed using streptavidin-HRP conjugated compact polymer system and visualized with chromogen substrate, 3’3-diamino-benzidine substrate (DAB). ![Immunocytochemistry/Immunofluorescence: IL-6 Antibody (1-6) [NBP2-89149] - Analysis of EL4 cells labeling IL-6 with Rabbit anti-IL-6 antibody at 1/1,000 dilution.Cells were fixed in 4% paraformaldehyde for 20 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 5 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-IL-6 antibody at 1/1,000 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (iFluor™ 488,) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI.Beta tubulin (red) was stained at 1/100 dilution overnight at +4℃.
