Rainbow Calibration Particles are used to ascertain the relative voltage range for each flow cytometer detector. This mixture of fluorescent particles serves as a reliable and reproducible means of evaluating the sensitivity and resolution of flow cytometers.
The relative number of fluorophores can been determined for every emission peak of the rainbow calibration particles in FL1 (FITC, MEFL), FL2 (RPE, MEPE), FL3 (RPE-Cy5, MEPCY) and FL4 (APC, MEAP) channels in order to plot the calibration graph. The calibration graph is used to check the linearity (dynamic range) of the photomultiplier tube (PMT) in each channel. Additionally, the relative number of fluorophores can be cross calibrated with cells or particles stained with a known number of spectral matching fluorophores such as FITC, PE, RPE-Cy5 to estimate the number of fluorophores on stained cells.
