Description

Regulated intramembrane proteolysis by '-secretase cleaves proteins in their transmembrane domain and is involved in important signaling pathways. gamma-Secretase catalyzes the intramembrane proteolysis of Notch, beta-amyloid precursor protein, and other substrates as part of a new signaling paradigm and as a key step in the pathogenesis of Alzheimer's disease. The intra-membrane proteolysis of amyloid-beta precursor protein (APP) and Notch along with many other cellular proteins is mediated by gamma-secreatase complex which is primarily composed of presenilin and nicastrin. The gamma secretase complex is formed by direct physical interaction of 4 proteins to form heterodimers including presenlin-2 (PEN-2), highly glycosylated nicastin, and mammalian Aph-1 that are genetically linked to gamma secretase. Coexpression of all 4 protein leads to a marked increase in presenilin heterodimers, fully glycosylation of nicastrin and enhanced gamma secretase activity (1). This Aph1 plays an important role in initial assembly and maturation of presenilin, nicastrin complexes. The gamma secretase complex plays a role in Alzheimer's disease and cancer progression. Specific inactivation of Aph1b in mouse Alzhiemer's model showed improvement in Alzhiemer's disease relevant phenotypic features without any Notch related side defects. Mammalian and C. elegans Aph1b is a very conserved protein, comprised of 257 amino acid with 7 TMD located on chromosome 15p22.2. The Aph1b protein is expressed in brain, hemopoetic cells and in many types of cancer cells and is invliced in protein trafficking, calcium regulation and apoptosis. The antibody also labels the anterior pharynx defective 1 homolog B in C. elegans. The Aph1b-selective antibodies were generated against unique antigenic sequences form near C-terminal end of the Aph1b protein. The antibodies to Aph1b are affinity purified over immobilized antigen based chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer.
