Western Blot: IL-1 beta/IL-1F2 Antibody [NB600-633] - Antibody was used at a 1:200 dilution incubated 1h at room temperature to detect dog IL-1b by Western blot. Lane 1 and 2 were loaded with 2.5 ug and 500 ng of dog IL-1b respectively. The molecular weight of the detected band is estimated by comparison to molecular weight markers (not shown). Detection occurred using a 1:3,000 dilution of IRDye (TM) 800 conjugated Donkey anti-Rabbit IgG for 1h at room temperature.

Effects of butyrate treatment on curdlan-induced SpA mice. SKG mice were injected with 3 mg of curdlan through intraperitoneal and oral administration. One week later, sodium butyrate (100 mg/kg) was administered orally once a day for 14 weeks. (A) Arthritis score and incidence were monitored by observation twice a week. Immunohistochemistry: TLR7 Antibody [NBP2-24906] - Expression of the TLRs 5 and 7 were evaluated in the PDAC tissue by their staining intensity using TLR5 antibody (NBP2-24787) and TLR7 antibody (NBP2-24906). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (84540, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: BLIMP1/PRDM1 Antibody (3H2-E8) [NB600-235] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-BLIMP1/PRDM1 (3H2-E8) conjugated to DyLight 550 [NB600-235R] at 20ug/ml for 1 hour at room temperature. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

The cells were incubated with anti- NBP1-76769 at 1 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Western Blot: PD-L1 Antibody - BSA Free [NBP1-76769] - Upregulated expression of PD‐L1 in cisplatin‐resistant cells through IL‐6/STAT3. The cisplatin resistant CAL27cis & Detroit‐562cis cells had increased IL‐6 mRNA & IL‐6 protein production compared to parental cells by qRT‐PCR & ELISA (A, B). CAL27cis & Detroit‐562cis were cultured with IL‐6 (10 ng/mL, human recombinant IL‐6, R&D Systems, Minneapolis, USA) for 48 h (C). Primary antibody was diluted 1:100 and incubated with samples for 24 hours at 4C. HRP-conjugated goat anti-rabbit antibody was used as the secondary antibody.

Western Blot: TLR3 Antibody [NB100-56571] - A TLR3 agonist polyinosinic‐polycytidylic acid (poly(I:C)) induced autophagy in cultured cardiomyocytes through a TRIF‐dependent pathway. (A) Poly(I:C) increased autophagy markers in cultured H9c2 rat ventricular cells. (B) Poly(I:C) stimulated autophagosome formation but did not affect autophagic flux. Primary cultured neonatal rat ventricular myocytes (NRVMs) were transfected with a tandem mRFP‐GFP‐LC3 adenovirus for 24 hrs, followed by treatment with poly(I:C) (100 μg/ml, 4 hrs). Autophagosomes & autolysosomes were, respectively, visualized as yellow‐ & red‐only punctas under a confocal microscope. (C) An autophagic flux inhibitor chloroquine (CQ) induced accumulations of LC3‐II & p62/SQSTM1 proteins in H9c2 myocytes receiving poly(I:C) (100 μg/ml, 4 hrs). CQ was applied at 10 μM, immediately prior to poly(I:C). (D) Effects of indicated siRNA on poly(I:C)‐induced changes in autophagy markers in NRVMs. All the siRNAs were transfected at 50 nM for 48 hrs, & poly(I:C) was added at 100 μg/ml for 4 hrs before cell harvest. Western Blot: RelA/NFkB p65 Antibody (112A1021) [NB100-56712] - Lysates of HeLa human cervical epithelial carcinoma cell line and Daudi human Burkitt's lymphoma cell line. PVDF membrane was probed with 1 ug/mL mouse anti-RelA/NFkB p65 monoclonal (NB100-56712, Novus Biologicals), followed by 1:2000 dilution of the appropriate HRP-conjugated secondary antibody, donkey anti-mouse IgG (HAF018).

Immunohistochemistry-Paraffin: RelA/NFkB p65 Antibody (112A1021) [NB100-56712] - Ovarian cystadenocarcinoma probed with p65 antibody at 5 ug/mL. Human tissue TMA was used for this test. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.

Flow Cytometry: RelA/NFkB p65 Antibody (112A1021) [NB100-56712] - Intracellular staining of 293 HEK cells using 0.5 ug of p65 antibody. Green histogram represents the isotype control (p65) antibody.
