![Analysis of cell line lysates probed with mouse mAb to GAPDH (theoretical molecular weight: 36 kDa), NB300-221, dilution 1:2,000: [1] protein standard, [2] HEK293, [3] HeLa, [4] SH-SY5Y, [5] COS1, [6] NIH-3T3, and [7] C6 cells. The GAPDH antibody reveals a single band at ~37 kDa in all cell lines. Bar graphs are ratios of optical densities of our proteins of interest to beta -actin (42 kDa band), expressed as immunoreactivity in percentage of the mean value of the WT group (100%). CD11b was significantly decreased in Munc18-OE mice compared to the WT (t = 3.01; P <0.05). No differences between groups were observed in the levels of GFAP or NF-kappa B. ![Western Blot: ATM Antibody (2C1) [NB100-309] - Rac1 inhibition abolishes IR-induced activation of both ATM & ATR signaling. (A) MCF-7 cells were treated with/without 20-Gy IR in the presence or absence of 100 μM NSC23766 & incubated for 1 hour at 37°C before analysis. To assess the ATM kinase activity, ATM was immunoprecipitated from cell lysate by using anti-ATM antibody (2C1) & assayed for ATM activity by using p53 recombinant protein as substrate. To measure the Chk2 activity, Chk2 was immunoprecipitated from cell lysate by using B-4 anti-Chk2 antibody & assayed for Chk2 activity by using Cdc25C recombinant protein as substrate. As controls, ATM & Chk2 protein levels in the immunoprecipitates (IP-WB) as well as in cell lysates (WB) were assessed with immunoblotting. (B) ATR & Chk1 were immunoprecipitated from cell lysates by using N-19 anti-ATR & G-4 anti-Chk1 antibody, respectively. ![Western Blot: mCherry Antibody (1C51) [NBP1-96752] - Threonine 276 is required for the intramolecular interaction, and for inhibition of membrane binding. Mutation of threonine 276 to aspartic acid promotes the association of cytohesin 2 with membranes. MDCK cells were transfected with constructs encoding mCherry-tagged wild-type or T276D cytohesin 2 and fractionated into cytosol and total membranes. The fractions were Western blotted with mouse anti-mCherry, mouse anti-E-cadherin and mouse anti-actin. ![Chromatin Immunoprecipitation: Histone H3 Antibody [NB500-171] - Chromatin from one million formaldehyde cross-linked HeLa cells was precipitated using 2 ug of NB500-171 and 25 uL of magnetic IgG beads, using standard ChIP methods. A similar sample containing no antibody (No Ab) was included as a negative control. Immunoprecipitated DNA was quantified using quantitative real-time PCR and SYBR green dye, then normalized to the non-precipitated input chromatin. The cells were incubated with anti CD68 (FA-11) [NBP2-33337] at a 1:100 dilution overnight at 4C and detected with an anti-rat DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue).
