SOX2 Antibody was detected in immersion fixed paraffin-embedded sections of human Glioblastoma using Mouse Anti-Human SOX2, Monoclonal Antibody (Catalog # MAB2018) at 5ug/mL at 37 ° Celsius for 4 minutes. Before incubation with the primary antibody, tissue underwent an all-in-one dewaxing and antigen retrieval preprocessing using PreTreatment Module (PT Module) and Dewax and HIER Buffer H (pH 9; Epredia Catalog # TA-999-DHBH). Tissue was stained using the Alexa Fluor™ 647 Goat anti-Mouse IgG Secondary Antibody at 1:200 at 37 ° Celsius for 2 minutes. (Yellow; Lunaphore Catalog # DR647MS) and counterstained with DAPI (blue; Lunaphore Catalog # DR100). Specific staining was localized to the nucleus. Protocol available in COMET™ Panel Builder.

E-Cadherin and SOX2 were detected in BG01V human embryonic stem cells using 10 µg/mL Goat Anti-Human E-Cadherin Antigen Affinity-purified Polyclonal Antibody (AF648) and 10 µg/mL Mouse Anti-Human/Mouse/Rat SOX2 Monoclonal Antibody (Catalog # MAB2018). Cells were incubated with primary antibodies for 3 hours at room temperature. Cells were stained for E-Cadherin using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; NL001) and for SOX2 using the NorthernLights 493-conjugated Anti-Mouse Secondary Antibody (green; NL009). Western blot shows lysates of NTera-2 human testicular embryonic carcinoma cell line, F9 mouse teratocarcinoma stem cells, D3 mouse embryonic stem cell line, and rat cortical stem cells. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human/Mouse/Rat SOX2 Monoclonal Antibody (Catalog # MAB2018) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (HAF018). A specific band was detected for SOX2 at approximately 36 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

NTera-2 human testicular embryonic carcinoma cell line was stained with Mouse Anti-Human/ Mouse/Rat SOX2 Monoclonal Antibody (Catalog # MAB2018, filled histogram) or isotype control antibody (MAB003, open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG Secondary Antibody (F0102B). Simple Western lane view shows lysates of BG01V human embryonic stem cells, loaded at 0.2 mg/mL. A specific band was detected for SOX2 at approximately 50 kDa (as indicated) using 4 µg/mL of Mouse Anti-Human/Mouse/Rat SOX2 Monoclonal Antibody (Catalog # MAB2018). Adjacent tissue section was processed for immunohistochemistry using R&D Systems Mouse Anti-Human CXCL12/SDF-1a Monoclonal Antibody (Catalog # MAB350) at 5 ug/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte HRP Polymer Antibody (R&D Systems, Catalog # VC001) and DAB chromogen (lower image, yellow-brown). Immunohistochemistry-Paraffin: Collagen IV Antibody [NB120-6586] - Collagen IV antibody at 1:400, 45 min RT showed strong staining in FFPE sections of human kidney (left) with strong red staining observed in glomeruli and liver (right) with strong staining in sinusoids. Staining for both tissues was consistent with a basement membrane distribution. Slides were steamed in 0.01 M sodium citrate buffer, pH 6.0 at 100C for 20 minutes for antigen retrieval.

A172 human glioblastoma cell line was stained with Rat Anti-Human/Mouse/Rat Vimentin Monoclonal Antibody (Catalog # MAB2105, filled histogram) or isotype control antibody (Catalog # MAB006, open histogram) followed by anti-Rat IgG PE-conjugated secondary antibody (Catalog # F0105B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Simple Western lane view shows lysates of Jurkat human acute T cell leukemia cell line, loaded at 0.2 mg/mL. A specific band was detected for Vimentin at approximately 58 kDa (as indicated) using 10 µg/mL of Rat Anti-Human/Mouse/Rat Vimentin Monoclonal Antibody (Catalog # MAB2105) followed by 1:50 dilution of HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

<b>Genetic Strategies Validation. </b>Western blot shows lysates of K562 human chronic myelogenous leukemia parental cell line and Vimentin knockout K562 cell line (KO). PVDF membrane was probed with 2 µg/mL of Rat Anti-Human/Mouse/Rat Vimentin Monoclonal Antibody (Catalog # MAB2105) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF017). The cells were incubated with anti-SOX2 (NB110-37235) at a 1:200 dilution overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin was used as a co-stain at a 1:1000 dilution and detected with and anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: SOX2 Antibody - BSA Free [NB110-37235] - NIH3T3 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with SOX2 Antibody (NB110-37235) at 2ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:1000 dilution for 60 minutes. Peripheral Node Addressin was detected in immersion fixed paraffin-embedded sections of human Tonsil using Rat Anti-Human Peripheral Node Addressin (MECA-79R), Monoclonal Antibody (Catalog #NBP2-78792) at 1:50 dilution at 37° Celsius for 4 minutes. Before incubation with the primary antibody, tissue underwent an all-in-one dewaxing and antigen retrieval preprocessing using PreTreatment Module (PT Module) and Dewax and HIER Buffer H (pH 9; Epredia Catalog # TA-999-DHBH). Tissue was stained using the Alexa Fluor™ 647 Goat anti-Rat IgG Secondary Antibody at 1:200 at 37 ° Celsius for 2 minutes. (Yellow; Lunaphore Catalog # DR647RT) and counterstained with DAPI (blue; Lunaphore Catalog # DR100). Specific staining was localized to endothelial cells. Immunohistochemistry-Paraffin: Peripheral Node Addressin Antibody (MECA-79R) [NBP2-78792] - Analysis of FFPE mouse adipose tissue section (with lymph node areas) using Peripheral Node Addressin antibody (clone MECA-79) at 1:100. The staining was developed with HRP-DAB detection method and the counterstaining was performed using hematoxylin. This Peripheral Node Addressin antibody generated a strong and specific staining of MECA-79 antigen in the the cytoplasm and the membranes of high endothelial venules (HEVs) aka peripheral lymph node addressin (PNAd) in lymph node areas of tested adipose tissue section.

Mouse splenocytes were stained with Rat Anti-Mouse CD4 APC-conjugated Monoclonal Antibody (Catalog # FAB554A) and either (A) Rabbit Anti-Human/Mouse FoxP3 Monoclonal Antibody (Catalog # MAB8214) or (B) Normal Rabbit IgG Control (Catalog # AB-105-C) followed by Phycoerythrin-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # F0110).
