On the next day, the cells were washed twice with PBS & incubated with the same compounds [solvent vehicle control (Veh; ≤0.1% DMSO), evodiamine (1–20 μM), & the PPAR gamma agonist pioglitazone (10 μM) as positive control] with or without 10 µg/mL apo A1. Extracellular as well as intracellular radioactivities were quantified with scintillation counter. At 72 hours post-transfection cells were chilled to 4 degrees C on ice and rVSV-SFTSV was bound by centrifugation (1200xg, 30', 4 degrees C). Following centrifugation, media was replaced with pre-warmed media (37 degrees C) and the cells placed in a 37 degrees C incubator for 40 minutes before fixation in 1% paraformaldehyde for 15 minutes. For each experiment, 500ul of DDK tagged CRYAB overexpression lysates (at 1:5 dilution with HEK293T lysate), 2ug of anti-CRYAB antibody and 20ul (0.1mg) of goat anti-mouse conjugated magnetic beads were mixed and incubated overnight.
