![Western Blot: Survivin Antibody (60.11) - BSA Free [NB500-238] - CD133+ cells are more resistant to chemotherapeutic agents. STA-ET-8.2 cells were FACS-sorted & CD133+ & CD133- fractions treated with increasing concentrations of (A) Doxorubicin (Doxo), (B) Etoposide (Etop), (C) Vincristine (Vinc) or (D) a combination of all three drugs at high (Doxo 10 μM; Etop 10 ug/ml; Vinc 100 ng/ml), medium (Doxo 2.5 μM; 20 Etop 2.5 ug/ml; Vinc 25 ng/ml) & low (Doxo 0.5 μM; Etop 0.5 ug/ml; Vinc5 ng/ml) concentrations. ![Immunohistochemistry-Paraffin: TRAILR2/TNFRSF10B Antibody [NB100-56618] - TRAIL R2/TNFRSF10B Antibody [NB100-56618] - Analysis of a formalin fixed paraffin-embedded (FFPE) human brain cerebellum using 1:200 conc. of TRAIL R2/TNFRSF10B antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 30 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 9.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 15 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. ![Immunocytochemistry/Immunofluorescence: IkB-alpha Antibody (6A920) [NB100-56507] - Hek293 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-IkB-alpha (6A920) at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). ![<b>Biological Strategies Validation. </b>Western Blot: IkB-alpha Antibody (6A920) [NB100-56507] - Jurkat cells were treated for 30 min with 100 ug/ml ALLN (N-Acetyl-Leu-Leu-Norleucinal; a Calpain inhibitor and also proteasome inhibitor that prevents IkBa dephosphorylation) followed by incubation with (lanes 2 & 4) or without 1 nM TNF-a (1 & 3). The membranes were blotted with NB100-56724 (lanes 1 & 2) or NB100-56507 (that recognizes both non-phospho and phosphorylated forms of IkBa) and immunoreactivity was detected by ECL.
