Western Blot: GAPDH Antibody [NB300-320] - stai Mutants Have Reduced Levels of alpha-Tubulin, Acetylated alpha-Tubulin, and Kinesin Heavy Chain Protein. Representative western blots of total protein extracted from stai mutant third instar larvae. Heterozygous stai/+ mutant larvae exhibit mild reductions in the levels of alpha-tubulin and acetylated alpha-tubulin compared with wild type larvae. Homozygous stai mutant larvae have dramatic reductions in the levels of alpha-tubulin, acetylated alpha-tubulin and the heavy chain subunit of the microtubule motor protein kinesin compared to wildtype. Df(2L) BSC239 represents a second chromosomal deficiency that excludes the stai gene. GAPDH is used as a loading control. Immunocytochemistry/Immunofluorescence: IRE1 alpha Antibody [NB100-2324] - IRE-1 alpha antibody was tested in HeLa cells at 1:200 against Dylight 488 (Green). Alpha tubulin and nuclei were counterstained against Dylight 568 (Red) and DAPI (Blue).

Immunohistochemistry-Paraffin: IRE1 alpha Antibody [NB100-2324] - Analysis of a formalin fixed mouse colon tissue section using total IRE1 alpha antibody (NB100-2324) at 1:300. Note a strong cytoplasmic signal in various cells of colonic villi (columnar epithelial and goblet cells) and the muscular layer. A few cells, potentially with active UPR, showed nuclear positivity also while the signal was negligible in cells at base of crypts and lamina propria.

Flow (Intracellular): Cytochrome c Antibody (7H8.2C12) [NB100-56503] - An intracellular stain was performed on HeLa cells with Cytochrome c (7H8.2C12) antibody NB100-56503PE (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to phycoerythrin.

Western Blot: Cytochrome c Antibody (7H8.2C12) [NB100-56503] - Human HeLa lysate probed with Cytochrome C antibody at 0.1 ug/ml.

Western Blot: Cytochrome c Antibody (7H8.2C12) [NB100-56503] - Analysis using the biotin conjugate of NB100-56503. Detection of Cytochrome C in 15 ug of HeLa cell lysate using NB100-55775 at 1:1000. A 15 kDa band is detected.

Flow Cytometry: Cytochrome c Antibody (7H8.2C12) [NB100-56503] - Using the Alexa Fluor 488 direct conjugate, an intracellular stain was performed on HeLa cells with Cytochrome c (7H8.2C12) antibody NB100-56503AF488 (blue) and a matched isotype control NBP2-27231AF488 (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Simple Western: Cytochrome c Antibody (7H8.2C12) [NB100-56503] - Lane view shows lysates of human heart tissue, loaded at 0.2 mg/mL. A specific band was detected for Cytochrome c at approximately 23 kDa (as indicated) using 2.5 ug/mL of Mouse Anti-Cytochrome c Monoclonal Antibody (Catalog # NB100-56503). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Immunohistochemistry-Paraffin: p73 Antibody (5B429) [NBP2-24737] - FFPE breast cancer probed with p73 antibody at 5 ug/ml. Human tissue was used for this test. Staining of tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.

Simple Western: p73 Antibody (5B429) [NBP2-24737] - Lane view shows a specific band for p73 in 1.0 mg/ml of HeLa lysate. This experiment was performed under reducing conditions using the 12-230kDa separation system. * Non-specific interaction with the 230 kDa Simple Western standard may be seen with this antibody.

<b>Biological Strategies Validation. </b>Immunohistochemistry: ASC/TMS1 Antibody [NBP1-78977] - Rats were infected by S. pneumoniae or given PBS sham control. 21.5 umoles/kg Mito-Vit-E (MVE) or vehicle was administered orally 30 minutes post-inoculation, and heart tissues were harvested 24 hours later. Heart sections were co-stained with anti-ASC (brown) and haematoxylin (blue). Negative control was stained with secondary antibody alone. Western Blot: ASC/TMS1 Antibody [NBP1-78977] - Whole cell protein from (1) THP1 and (2) HL60 was separated on a 4-15% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 4.0 ug/ml anti-ACS/TMS1 in 1% milk, and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.

Flow Cytometry: ASC/TMS1 Antibody - BSA Free [NBP1-78977] - An intracellular stain was performed on MCF7 cells with ASC/TMS1 NBP1-78977 (blue) and a matched isotype control NBP2-24891 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: ASC/TMS1 Antibody [NBP1-78977] - Tested in MCF-7 cells with FITC (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and DyLight 550 (red).

Effects of abrocitinib on NF kappa B-related inflammation and pyroptosis pathways. On the 1st day after TBI, NF kappa B-related inflammation and the activation of pyroptosis pathways were dramatically increased. After abrocitinib treatment, the indicators of NF kappa B-related inflammation and pyroptosis pathways were significantly decreased, as can be seen from the WB (n = 5–9 one-way ANOVA with Tukey’s post-hoc test) and GSDMD IHC staining (A–C) (n = 6 Kruskal-Wallis test). $Western Blot: GAPDH Antibody [NB300-327] - Analysis of different cell cytosolic or nuclear enriched fractions, GAPDH antibody, dilution 1:20,000 (Red): [1] protein standard, [2] NIH-3T3 cytosolic, [3] NIH-3T3 nuclear, [4] HeLa cytosolic, and [5] HeLa nuclear fractions. Strong band at 37kDa corresponds to GAPDH protein, mainly detected in the cytosolic fractions. The same blot was simultaneously probed with mouse mAb to the nuclear RNA binding protein SF3B4, dilution 1:1,000 (Green). In contrast to GAPDH, SF3B4 is exclusively expressed in the nuclear fraction.$

The cells were incubated with anti-HIF Prolyl Hydroxylase 3 (NB100-3030) at 1:200 overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at 1:500. Alpha tubulin, DM1A (NB100-690) was used as a co-stain at 1:1000 and detected with an anti-mouse DyLight 550 (Red) at 1:500. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Western Blot: OPA1 Antibody [NB110-55290] - Total protein from Human HeLa cells, Mouse MEF cells and Rat Brain was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-OPA1 in 1% blocking buffer. Precision Plus Protein All Blue molecular weight markers (BioRad) were detected with 1 ug/ml Anti-Blue Marker antibody (NBP2-33376). The protein ladder was detected with an anti-mouse HRP secondary antibody and OPA1 with an anti-rabbit HRP secondary antibody using chemiluminescence.

Flow Cytometry: p53 Antibody (PAb 240) [NB200-103] - An intracellular stain was performed on HeLa cells with p53 Antibody [PAb 240] NB200-103 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (35503, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: p53 Antibody (PAb 240) [NB200-103] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-p53 (PAb 240) [NB200-103] at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: p53 Antibody (PAb 240) [NB200-103] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-p53 Antibody (PAb 240) NB200-103 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunocytochemistry/Immunofluorescence: p53 Antibody (PAb 240) [NB200-103] - Neuro2a cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by APC-conjugated anti-mouse IgG secondary antibody F0101B.

The effect of the co-culturing of cancer cells and fibroblasts on the expression of Hk2 and Ldha.
