Th-POK Antibody Summary

This antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an internal sequence of mouse Th-POK protein.
This antibody is directed against mouse cKrox. A BLAST analysis was used to suggest cross-reactivity with c-Krox from mouse, human and rat based on a 100% identity with the immunizing sequence.

Application Notes
Expect a band approximately 80 kDa in size corresponding - cKrox by western blotting in the appropriate cell lysate or extract. Lysates from mouse thymocytes are recommended for use as positive controls.

cKrox (also known as Zfp67 or ThPOK) functions as a transcription regulator that acts as a key modulator of lineage commitment for immature T-cell precursors. cKrox is necessary and sufficient for commitment of CD4 lineage, while its absence causes CD8 commitment. Development of immature T-cell precursors (thymocytes) to either the CD4 helper or CD8 killer T-cell lineages correlates precisely with their T-cell receptor specificity for major histocompatibility complex class II or class I molecules, respectively. cKrox is a known transcriptional repressor of the collagen COL1A1 and COL1A2 genes and may also function as a repressor of fibronectin and possibly other extracellular matrix genes. cKrox is located within the nucleus and is expressed in multiple tissues and organs including skin, thymus and peripheral T-cells.

  1. Can you send me the validation data for the IHC-P or put it on the website and what ab dilution or amount is required for that application? Are you sending me in house validation images, or putting them on the website?
    • We recommend a concentration of 1.25-2.5 ug/ml for use of NBP1-77810 in IHC on paraffin-embedded tissues. If we have images, they will be posted on the website. This may have been validated for IHC-P use by customer feedback so images may not be available. It is still guaranteed for all applications and species listed on our datasheet.
