Application Notes
Western Blot - Sample preparation: Resuspend approx. 50 mil. cells in 1 ml cold Lysis buffer (1% laurylmaltoside in 20 mM Tris/Cl, 100 mM NaCl pH 8.2, 50 mM NaF including Protease inhibitor Cocktail). Incubate 60 min on ice. Centrifuge to remove cell debris. Mix lysate with non-reducing SDS-PAGE sample buffer. The antibody MEM-01 stains only the zone corresponding to 140 kDa, but not the weaker 130 kDa zone (stained by several CD22 antibodies recognizing both isoforms).

Background

The antibody MEM-01 reacts with CD22 (BL-CAM), a 130 kDa type I transmembrane glycoprotein (immunoglobulin superfamily) expressed in the cytoplasm of pro-B and pre-B lymphocytes, and on the surface of mature and activated B lymphocytes; it is lost on plasma cells, peripheral blood T lymphocytes, granulocytes and monocytes. The antibody MEM-01 cross-blocks the antibody OTH228 that recognizes uniquely epitope "E"; it does not cross-block NB100-78574.

  1. I am interested in buying a CD22 antibody for fluorescence microscopy and FACS I red on your web page that you also offer to label your antibodies, if required, is that correct?
    • We do have the ability to perform custom conjugations on certain antibodies, including some to the CD22 protein. If you send me the conjugate you are interested in I can provide you with a quote.
  2. Just a few things to clarify. Firstly, will the biotin binding sites on the streptavidin be available after conjugation (at least some of them)? Secondly, presumably the Antibody-streptavidin conjugation procedure is not 100% efficient (i.e. not all antibodies will be conjugated to streptavidin). So will you send me the purified Antibody-streptaviding complex (without the unconjugated Antibody)? And does that in turn mean that will receive not the full amount of the NB500-421 antibody as quoted on your website, but a proportion thereof?
    • The Biotin binding sites should be available on the conjugate for your experiments. The kit we use to conjugate is designed to have a 4:1 ratio of conjugate to antibody so theoretically it should conjugate the entire amount located in the vial since they bind to any free amines. However, when doing a custom conjugation we cannot guarantee yield after the conjugation the lab has informed me. Typically the recovery will be high and you should have close to the amount listed on the unconjugated antibodies datasheet but when conjugating a single unit we cannot perform any quantitative analysis as this analysis typically loses about 30% of the antibody. Also note that we do have streptavidin conjugation kits that you can purchase to perform the conjugation yourself if you are worried about recovery and would prefer to perform this yourself.
  3. can your lab tell me approximately what minimum coupling efficiency I can expect (maybe from past experience)? I just want to get a rough idea. Since you are using an excess of streptavidin, will there be free streptavidin in my sample (which will compete with my biotin ligand), or is the excess streptavidin removed?
    • Typically the minimum coupling efficiency would be 1:1 from past experience. All of the excess unbound streptavidin should be quenched after the conjugation so there will be no competition for signal with your biotin.
