Background

Recoverin (RCVRN; also called Protein CAR), is a cytoplasmic calcium-binding protein which is found mainly in the pineal gland and retina, distributed in photoreceptor cells, bipolar cells, as well as in a small population of cells in ganglion cell layer. In photoreceptor cells, this neuronal calcium sensor protein plays a key role in the recovery phase of phototransduction by regulating activity of rhodopsin kinase (RK). Recoverin is consists of two domains each containing a pair of EF-hand-type Ca2+-binding sites that binds two Ca2+ ions: one with high affinity, and the other with low affinity. Upon binding of Ca2+ ions, recoverin undergoes a series of conformational changes allowing the exposure of amino acid residues of its hydrophobic pocket intended for interaction with RK. In parallel, the myristoyl group of recoverin (N-terminus acylated with myristic acid) becomes exposed imparting the interaction with photoreceptor membranes (so-called Ca2+/myristoyl switch mechanism). In photoreceptor cells, recoverin inhibits RK in dark while this inhibition is relieved upon illumination, and the subsequent drop of intracellular Ca2+ levels. Recoverin allows RK to phosphorylate bleached receptor, rhodopsin, which is the first step to its desensitization.
