This RBFOX3/Neun (1B7) antibody is useful for ICC and Western blot, where bands can be seen at 46 and 48 kDa. Use in IHC-frozen sections was reported in the scientific literature (PMID: 23776455). RBFOX3/NeuN (RNA binding protein FOX-1 homolog 3/ Neuronal nuclei) is a RNA-binding protein localized in the nucleus as well as cytoplasm that regulates alternative splicing events. It is a member of RNA-binding FOX (RBFOX) family of splicing regulators which comprises RBFOX1 (Fox-1 or A2BP1), RBFOX2 (Fox-2 or RBM9), and RBFOX3 (Fox-3, HRNBP3, or NeuN). All of these Fox protein have single central RNA recognition motif RNA-binding domain which detects sequence (U)GCAUG observed within introns flanking alternative exons. (U)GCAUG motif position corresponding to the alternative exon dictates RBFOX's effects on splicing process. A motif localized upstream from alternative exon generally repress RBFOX-dependent exon inclusion, whereas, a downstream stream motif will usually promote inclusion. Neurons, heart, and muscle express RBFOX 1, whereas, RBFOX3 is limited to neurons only, and RBFOX2 is expressed in these tissues alongwith expression in embryos, hematopoietic cells, and embryonic stem cells. Defects in alternative splicing are known to result in neurological and neuromuscular diseases such as frontotemporal dementia and myotonic dystrophy.
