Ran is a small GTPase of the Ras superfamily that controls nucleocytoplasmic transport. However, Ran also functions in the absence of a membrane-enclosed nucleus, during cell division. RanGTP acts to promote microtubule nucleation during mitotic spindle assembly and, later during mitosis, is required for NE assembly. For correct spindle and NE assembly on chromatin, it is critical that the RanGTP concentration on or near chromatin is
higher than in the surrounding mitotic cytoplasm. Unlike many other regulatory GTPases, Ran is not posttranslationally modified so that it can be targeted to the subcellular compartment where it acts. Rather, RanGTP has been thought to be localized by the differential localization of Ran regulators.
During mitosis, the nucleotide exchange factor RCC1, which generates RanGTP, is in part bound to chromatin. This results in the preferential production of RanGTP on the chromatin surface. From there, RanGTP is believed to diffuse only a short distance before either encountering mediators of spindle or NE assembly or, alternatively, being reconverted to RanGDP under the influence.

Our customer would like to purchase this antibody for IP (catalog # NB100-1048). They would like to have its concentration to estimate the titer for IP. Since it is the whole antisera product, we assume that there is no info of concentration. If so, would you please help suggest the dilution titer for IP with this antibody?
NB100-1048 is not purified so the exact concentration is not quantifiable. As far as the recommended dilution for IP is concerned, try 1:50 to 1:100 dilution as the starting point and if IP doesn't look very well, further optimization steps might be required.
