RAB7A (Ras-related protein Rab-7a), a member of small GTPase superfamily/Rab family, is widely expressed with highest expression levels in skeletal muscles and localize to late endosome, lysosome, phagosome, melanosome etc. It colocalizes with OSBPL1A at the late endosome and interacts with RAC1, NTRK1/TRKA, RILP, PSMA7, RNF115, FYCO1, OSBPL1A, CLN3, and PIK3C3/VPS34-PIK3R4 complex. RAB7A is a major player in endo-lysosomal trafficking and it directs the early-to-late endosomal maturation, microtubule minus/plus-end directed endosomal migration as well as positioning, and endosome-lysosome transport through different protein-protein interaction cascades. RAB7A regulates growth-factor-mediated cell signaling, nutrient-transportor mediated nutrient uptake, neurotrophin transport in neuronal axons, lipid metabolism, pathogen infections including viral life cycle and regulation of certain specialized endosomal membrane trafficking such as melanosomes maturation, pathogen-induced phagosomes, autophagosomes etc. With RAC1, RAB7A controls the ruffled borders formation in osteoclasts. It also regulate endosomal trafficking and neurite outgrowth signaling of NTRK1/TRKA, and endocytic trafficking of EGF-EGFR complex. Defects in RAB7A have been linked to Charcot-Marie-Tooth disease type 2B (CMT2B).
