PHLPP Antibody Summary
The immunogen recognized by this antibody maps to a region between residue 1150 and the C-terminus (residue 1205) of human PH Domain and Leucine Rich Repeat Protein Phosphatase using the numbering given in entry NP_919431.1 (GeneID 23239).
PHLPP antibody has been reported to cross reacts with beta-catenin, which may display 90kDa band. We would not recommend using this specific antibody for assays where signals may not be notable between PHLPP1 and Beta-Catenin.

Western Blot: Beta catenin band in Western blot is relatively less intense than PHLPP band. We do not recommend this antibody in Immunohistochemistry, Immunocytochemistry and Immunofluorescence due to its cross reactivity to Beta Catenin. Use in Immunocytochemistry/immunofluorescence and Immunohistochemistry-Paraffin reported in scientific literature (PMID 25820252)

PHLPP (PH domain leucine-rich repeat-containing protein phosphatase) is responsible for the dephosphorylation and subsequent inactivation of Akt. Akt is a signaling protein critical to the balance of cell survival and apoptosis. Dephosphorylation of Akt by PHLPP has been demonstrated to trigger apoptosis and suppress tumor cell growth. PHLPP functions similarly to its relative PHLPPL, also known as PHLPPL2. Recent studies show that PHLPP and PHLPPL selectively terminate Akt-signaling by targeting specific Akt isoforms. Alternate names for PHLPP include pleckstrin homology domain-containing family E protein 1, suprachiasmatic nucleus circadian oscillatory protein, hSCOP, PHLPP1, KIAA0606, PLEKHE1, and SCOP.
