Application Notes
The ATM2 antibody has been tested by flow cytometric analysis of activated rat splenocyte suspensions. This can be used at less than or equal to 0.5 ug per test. Cell number should be determined empirically but can range from 10^5to 10^8cells/test.

Background

The ATM2 monoclonal antibody reacts with rat OX40 Ligand, a member of the TNF superfamily. OX40L is constitutively expressed by splenic dendritic cells and can be upregulated on splenic B cells via stimulation with LPS. Expression of OX40L has not been detected on rat peritoneal macrophages, resting or activated T cells. The interaction of OX40 (CD134) with OX40L has been implicated in T cell-dependent humoral response. ATM2 inhibits the costimulatory activity of rat OX40L transfected cells for anti-CD3-stimulated T cell proliferation and cytokine production.
