NB600-1394 was tested in a sandwich ELISA along with HRP-conjugated NB600-1528 as the detection antibody. Please note that you will need to optimize the conditions according to your specific samples and equipment. 1-10 ug of NB600-1394 in PBS are added to wells of a microtiter plate and incubated overnight at 4C. The wells are then washed three times with PBS and blocked with non-fat dry milk solution at room temperature for 2 hours. This blocking solution is discarded and 100ul of sample is added. After 1 hour of incubation with slight agitation at room temperature, wells are washed three times with PBS and 100ul of NB600-1528 labeled with horseradish peroxidase are added to each well. After 1 hour of incubation with slight agitation at room temperature, unbound labeled antibody is removed by washing three times with PBS and 100ul of OPD substrate solution are added to each well. After 15-30 minutes of incubation, the reaction is stopped with 100ul HCl and absorbance is read at 492nm.
